Levels of bovine IgA, IgG1 and total protein (TP) were determined in serum, saliva, tears and individual quarter lacteal secretions of six Holstein-Friesian cows sampled from six weeks before to four weeks after parturition. Hierarchal analyses of variance indicated significant variations among weeks, cows and quarters of the udder. A precipitous but non proportional drop in the levels of IgA and IgA1 in lacteal secretions occurred at calving. There was a concomitant increase in IgG1, and decrease in IgA, in serum. Correlation studies supported the concept of selective transport of IgG1 from serum to lacteal secretions in regulated amounts independent of serum IgG1 levels. Changes in the IgG1/TP ratio of serum and lacteal secretions supported the idea of a decrease in the selective transport mechanism. Correlation studies and estimations of secretory IgA (SIgA) in serum suggest that serum IgA is derived from IgA synthesized in secretory tissues. Highly significant correlations between IgA and IgG1 levels in all secretions postpartum suggest that local IgA synthesis and either IgG1 transport or local IgG1 synthesis are initiated by the same stimuli. Although some of the variation in the level reported for IgA and IgG1 in secretions resulted from protein dilution, much of the variation represents physiological differences between individual animals and tissues in the same animal. An IgG2/IgG1 ratio approaching that of serum occurred in a mastitic quarter of one cow. IgA was the principal immunoglobulin in saliva and tears, comprised a greater proportion of the immunoglobulin in milk whey than in prepartum lacteal secretions and was a minor immunoglobulin in bovine serum.  相似文献

Effects of whole-tree clearcutting are being studied in three major forest types in the northeastern United States: a spruce-fir forest in central Maine, a northern hardwood forest in New Hampshire, and a central hardwood forest in Connecticut. At each site we sampled total and extractable nutrient capitals, inputs and outputs of nutrient ions in precipitation and streamflow, nutrient removals in harvested products, and nutrient accumulation in regrowth. Depending upon location, combined losses of nutrients in harvested products and increased leaching to streams were in the ranges of 374–558 kg ha−1 for Ca, 135–253 kg ha−1 for K, 50–65 kg ha−1 for Mg, 248–379 kg ha−1 for N, and 19–54 kg ha−1 for P. Opportunities for replacing these losses over the next rotation are best for N. Data on inputs in precipitation versus outputs in streamflow indicate that, once effects of harvest subside, most N in precipitation will stay within the forest. By contrast, Ca shows a net output of 8–15 kg ha−1 year−1 from uncut watersheds, and the added leaching losses due to harvest may have a serious impact on Ca capital. This is especially the case for the Connecticut site, where total site capital for Ca is only about 4000 kg ha−1.  相似文献

Summary A considerable amount of data has been collected over the past few years on the rupture life of particleboard. This information is particularly valuable at the present time because there is increasing interest in the use of particleboard as a structural material. This paper presents a conventional analysis of the time to failure of three commercially available brands of particleboard at 20°C and at three fixed levels of relative humidity. A linear relationship is assumed between the logarithm of time to failure and stress level, and an improvement is made in the usual method of analysis by including censored specimens. The regression lines are projected forward to give estimates of the stress levels that can be sustained for a particular design life. It is found that the regression lines for 30 per cent and 65 per cent rh may be combined, but the specimens at 90 per cent rh are significantly lower in strength at any given time. One of the MUF bonded boards performs significantly better than a second MUF board over the long term, and this gives added weight to the view that there are other factors in the make-up and production of the board that are at least as important as the resin type itself.  相似文献

Evolving agricultural policies have influenced management practices within agroecosystems, impacting available habitats for many species of wildlife. Enhancing wildlife habitat has become an explicit objective of existing agricultural policy. Thus, there is renewed focus on field borders and the use of shelterbelt agroforestry systems to achieve conservation goals in the Midwest. Two Representative Farms – a 283-ha dryland and 510-ha irrigated farm were created in Saunders County, Nebraska. The Habitat Analysis and Modeling System (HAMS) was used to describe the composition and spatial pattern of the existing farms and surrounding landscape, as well as for the landscapes surrounding selected Breeding Bird Survey (BBS) routes. Simulated land use changes resulting from the implementation of two shelterbelt scenarios, Agricultural and Wildlife, were incorporated on each Representative Farm and surrounding landscape. Landscape variables which influence breeding bird species richness and community composition as determined from BBS routes were measured on simulated farm landscapes. A more heterogeneous landscape results from implementing either scenario. The percent total woods was a significant determinant of bird species richness on the BBS routes and was important in influencing bird communities at the farm- and landscape-level. Other landscape metrics which influenced the bird community composition on BBS routes were woody edge percentages and edge density values. Policies promoting shelterbelts create edge habitats which ultimately favor birds within the Forest-edge/generalist guild while bird species in need of conservation such as grassland-field species would potentially be negatively affected. This study was conducted to identify and analyse the expression of gametogenesis‐associated genes and proteins in foetal and adult buffalo gonads of both the sexes. Relative quantification of the genes was determined by qPCR and Western blotting. Immunohistochemistry was also performed for various gametogenesis‐associated proteins in foetal and adult gonads of both the sexes. We observed significantly (p < 0.05) increased expression of primordial germ cell‐specific, meiotic as well as genes associated with oocyte maturation and development in foetal ovaries as compared to the adult ones. However, significantly (p < 0.05) increased expression of proteins associated with oocyte maturation like GDF9 and ZP4 was found in adult ovaries, indicating temporal regulation of mRNA translation during oogenesis. Meiotic genes showed significantly (p < 0.05) increased expression in adult testes as compared to foetal testes and ovaries, indicating onset of meiosis at a later stage in spermatogenesis. In general, the expression of primordial germ cell‐associated as well as meiotic genes was higher in adult testes, indicating the increased biological activity in the organ. Immunohistochemistry revealed localized expression of gametogenesis‐associated proteins in ovarian follicles and seminiferous tubules of testes, while the surrounding somatic tissues were devoid of these proteins. The study gives an understanding of the sequential and temporal events of gene expression as well as mRNA translation during male and female gametogenesis. It could also be concluded that follicles and seminiferous tubules are the functional units of the female and male gonads, respectively, and their function could be enhanced by appropriate chemical and genetic intervention of the somatic tissue immediately surrounding them. This assumes importance in the context that buffalo attains sexual maturity at an older age of 2–3 years and have smaller ovaries with lesser number of primordial follicles in comparison with cattle, which is suggested to be the main reason of their poor breeding performance.  相似文献

Detailed observations of the behaviour of harbour seals, Phoca vitulina L., at sites within the estuaries of the Rivers Dee and Don, in north-eastern Scotland, were made over two full years between 1993 and 1996. Small numbers of grey seals, Halichoerus grypus Fab., were also present. The presence of seals within the estuaries was strongly related to season, with maximum numbers observed in winter and early spring; seals were virtually absent in June and July. The River Don was used largely as a haul-out site, while the River Dee was used predominantly as a foraging site, although it was not possible to determine whether the same seals were using the two estuaries. More seals were hauled-out on the River Don during twilight and dark than in daylight. The seals were observed to eat mostly salmonids, Salmo salar L. and S. trutta L., unidentified roundfish and flounder, Pleuronectes flesus L. The otoliths identified in scats collected at the mouth of the River Don belonged to marine species indicating that the seals were also feeding outside the estuaries. A minimum estimate is given of the numbers of large salmonids eaten in each river during the course of the year. Although no information was available on the numbers of salmonids using the rivers or the reproductive status of the fish eaten by the seals, as a cause of mortality, seal predation on large salmonids in estuaries is apparently an order of magnitude less important than mortality caused by angling within the river.  相似文献

Vegetated drainages are an effective method for removal of pollutants associated with agricultural runoff. Leersia oryzoides, a plant common to agricultural ditches, may be particularly effective in remediation; however, research characterizing responses of L. oryzoides to flooding are limited. Soil reduction resulting from flooding can change availability of nutrients to plants via changes in chemical species (e.g., increasing solubility of Fe). Additionally, plant metabolic stresses resulting from reduced soils can decrease nutrient uptake and translocation. The objective of this study was to characterize belowground and aboveground nutrient allocation of L. oryzoides subjected to various soil moisture regimes. Treatments included: a well-watered and well-drained control; a continuously saturated treatment; a 48-h pulse-flood treatment; and a partially flooded treatment in which water level was maintained at 15 cm below the soil surface and flooded to the soil surface for 48 h once a week. Soil redox potential (Eh, mV) was measured periodically over the course of the 8-week experiment. At experiment termination, concentrations of Kjeldahl nitrogen, phosphorus (P), potassium (K), iron (Fe), and manganese (Mn) were measured in plant tissues. All flooded treatments demonstrated moderately reduced soil conditions (Eh < 350 mV). Plant Kjeldahl nitrogen concentrations demonstrated no treatment effect, whereas P and K concentrations decreased in aboveground portions of the plant. Belowground concentrations of P, Mn, and Fe were significantly higher in flooded plants, likely due to the increased solubility of these nutrients resulting from the reductive decomposition of metal–phosphate complexes in the soil and subsequent precipitation in the rhizosphere. These results indicate that wetland plants may indirectly affect P, Mn, and Fe concentrations in surface waters by altering local trends in soil oxidation–reduction chemistry.  相似文献

Ecology of Freshwater Fish 2010: 19: 361–372. © 2010 John Wiley & Sons A/S Abstract – Dams without fish passage facilities block access to much of the historic spawning habitat of spring Chinook salmon (Oncorhynchus tshawytscha) in Oregon’s Willamette River basin. Adult salmon are routinely outplanted above the dams to supplement natural production, but many die before spawning despite extensive suitable habitat. In 2004–2007, we examined prespawn mortality patterns using live detection and carcass recovery data for 242 radio‐tagged outplants. Total prespawn mortality was 48%, but variability was high, ranging from 0% to 93% for individual release groups. Prespawn mortality was strongly condition dependent, consistently higher for females than males and higher for early release groups. Across years, warm water temperature in the migration corridor and at the collection site was associated with sharply higher mortality. Results highlight a need for better evaluations of the effects of adult mortality on population reintroduction and recovery and relationships among prespawn mortality, dam‐related temperature change and salmon life history and behaviour.  相似文献
